Chronic inflammation can cause a decrease in NAD+

With age, NAD+ (nicotinamide-adenine-dinucleotide) a key metabolite essential to a healthy and efficient metabolism declines. This phenomenon, which was previously unknown, is linked to a number of age-related illnesses and has led to the development of nutritional supplements that aim at restoring NAD+ levels. Researchers at the Buck Institute published their findings in Nature Metabolism. They identified chronic inflammation to be a major driver of NAD+ depletion. Researchers at the Buck Institute have shown that an increase in senescent (aging) cells causes NAD degradation via activation of CD38, a protein found on cell membranes inside and outside of many immune cells.

The fact that chronic inflammation and NAD+ decline are linked provides a holistic, systemic view of aging. And the discovery of CD38 macrophages as the mediator between the two gives a new therapeutic target.
