The mitochondrion is a long-time fascination for biologists, and its diversity of targets makes it a promising area for drug development. However, little has been achieved in the clinic. The potential to improve patient health and reduce costs is enormous, given the variety of mitochondrial diseases and inborn errors of metabolic metabolism, as well as mitochondrially-mediated oxidative stresses (myopathies and reperfusion injuries, Parkinson’s Disease, and ageing). This special issue of British Journal of Pharmacology highlights the current advances in mitochondrial biology and the emerging drug targets, as well as the new molecules that are nearing clinical application. The contributions of the authors are diverse, both in terms species and organs studied and in terms of the mechanisms being studied. Updated definitions of mitochondrial roles in disease and descriptions of previously unknown contributions are presented in terms of the interface of basic science and disease relevance.
