In a human trial, nicotinamide-riboside supplementation improved vascular function. However, researchers here have shown that an increase in NAD+ is likely to worsen the inflammatory signals of senescent cell. Senescent cells are a major cause of chronic inflammation that is associated with aging.
The results suggest that senolytic treatments, which selectively destroy senescent cell populations in older people, should be used before any current methods of raising NAD+ levels. It is still unclear whether the available options will provide NAD+ enhancement in the long term with safety. People who are self-experimenting with NAD+ supplementation may want to keep an eye on inflammation markers.
